Protective effect of curcumin analogue L6H4 on diaphragm of type 2 diabetic rats

AIM: To investigate the protective effect of curcumin analogue L6H4 on diaphragm of type 2 diabetic rats.METHODS: SPF male Sprague-Dawley rats (n=40) were randomly divided into 5 groups: normal control (NC) group, high fat (HF) group, high fat+L6H4 treatment (FT) group, diabetes mellitus (DM) group and DM+L6H4 treatment (DT) group. The rats in the later 4 groups were fed with high-fat diet. After 4 weeks of high-fat diet fee-ding, the rats in DM and DT groups were intraperitoneally injected with streptozotocin to induce type 2 diabetes melliutus. The rats in FT and DT groups were given L6H4 by gavage for 8 weeks. Blood glucose and blood lipid levels were detected biochemically. Fasting serum insulin (FINS) level was measured by radioimmunoassay and insulin resistance index (HOMA-IR) was calculated. Serum adiponectin (APN) level was measured by ELISA. The morphological changes of the diaphragm were observed under light and transmission electron microscopes. Lipid deposition and the activity of succinate dehydrogenase (SDH) and NADH-tetrazolium reductase (NADH-TR) were observed by enzyme histochemical staining. The content of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in the diaphragm were measured by thiobarbituric acid method and hydroxylamine method, respectively. The protein expression of adiponectin receptor 1 (AdipoR1) in the diaphragm was determined by immunohistochemistry and Western blot. RESULTS: The levels of blood lipids, blood glucose, FINS and HOMA-IR in HF and DM groups were higher than those in NC group, but decreased after L6H4 treatment. The serum APN level in HF and DM groups was lower than that in NC group, but increased after treatment with L6H4. The muscle fibers of the diaphragm were shrunk, fat particles accumulated in the muscle fibers, and the mitochondria were slightly swollen in HF and DM groups. The diaphragmatic fibrosis was obvious in DM group. These lesions were relieved after L6H4 treatment. Compared with NC group, the level of MDA and the activity of SDH and NADH-TR in the diaphragm were increased in HF and DM groups, but decreased after treatment with L6H4. The activity of SOD and the expression of AdipoR1 in the diaphragm were lower than those in NC group, but increased after L6H4 treatment.CONCLUSION: The curcumin analogue L6H4 exerts a protective effect on diaphragm in type 2 diabetic rats. The strengthened protein expression of AdipoR1, the increased serum level of APN, and anti-lipid peroxidation may be involved in the process.

通过不同蔟具及蔟中环境对彩色茧品种秋丰×黄3茧丝质影响的分析,我们得出：在自然通风或封闭环境中,用方格蔟上蔟蚕茧的一茧丝长、解舒长、解舒率、净度和清洁度要比用蜈蚣蔟好,其中解舒率分别提高了6.6%、5%;用方格蔟或蜈蚣蔟上蔟,在自然通风的蔟中环境下所得到的蚕茧茧丝质量比在封闭环境下高,解舒率更是分别提高了11.4%、9...     

腐殖酸与EDTA对高铁酸钾降解水中孔雀石绿影响的研究

在高铁酸钾与孔雀石绿的质量比为10∶1,pH值为7.0的条件下,考察腐殖酸与EDTA对高铁酸钾降解水体中孔雀石绿的影响。结果表明,加入少量的腐殖酸和EDTA溶液,孔雀石绿溶液的吸光度降低,有利于高铁酸钾对孔雀石绿的降解;EDTA的加入量过多时,反而抑制高铁酸钾对孔雀石绿的降解;加入2.0 mL腐殖酸和4.0 mL EDTA溶液降解2 h效果较好。     

草莓FaSKP1-1基因的克隆与表达分析

为研究草莓中SCF复合体的功能,以栽培草莓品种‘74’为试材,采用同源克隆的方法分离出2个SKP1基因。这2个基因核苷酸序列全长均为519bp,核苷酸序列相似性为98.46%,氨基酸序列相似性为98.84%,并具有特殊的‘GVDED’尾巴结构,分别命名为FaSKP1-1a和FaSKP1-1b(基因登录号分别为KU975057和KU975058)。RT-PCR和dCAPS分析发现FaSKP1-1a和FaSKP1-1b在草莓根、茎、叶、花托、花粉、花柱、果实中均高表达,在花瓣中表达量低。上述结果表明FaSKP1-1可能在草莓SCF复合体的形成过程中发挥重要作用。     

15种杀虫剂对草地贪夜蛾卵的毒力测定

采用浸卵法测定了15种杀虫剂对草地贪夜蛾卵的毒力,并比较了不同杀虫剂的毒力水平。结果表明:100 mg/L处理后48 h,甲氨基阿维菌素苯甲酸盐(甲维盐)和灭多威对草地贪夜蛾卵孵化抑制率最高,达到100%,其LC 50分别为12.67 mg/L和16.95 mg/L;而200 mg/L的氯虫苯甲酰胺、多杀霉素、阿维菌素、高效氯氰菊酯、虱螨脲、辛硫磷和杀螟丹分别处理后72 h对草地贪夜蛾卵的孵化抑制率次之,在69.6%~87.65%之间,其LC 50分别为44.1、50.9、78.37、79.0、98.15、126.8 mg/L和137.7 mg/L。依据毒力测定的结果和高效低毒类杀虫剂优先选用的原则,推荐选择甲维盐、氯虫苯甲酰胺、多杀霉素、阿维菌素和高效氯氰菊酯作为防治草地贪夜蛾卵的主要药剂。     

阿魏酸对2种野生杜鹃种子萌发的化感作用

为探究化感物质阿魏酸对马缨杜鹃和露珠杜鹃种子萌发的化感作用,本研究采用不同浓度阿魏酸分别处理马缨杜鹃和露珠杜鹃种子,利用单因素方差对其发芽率、发芽势、发芽指数和化感指数进行差异性分析.结果表明:阿魏酸显著抑制马缨杜鹃和露珠杜鹃种子的萌发,对2种杜鹃种子的抑制作用随浓度的增加而增加.阿魏酸对马缨杜鹃和露珠杜鹃种子均具有显著的化感作用,化感效应的最强浓度为1 000 μg/mL.     

Effect of Psammaplin A on in vitro Development of Bovine Aging Oocytes

To investigate the effect of histone deacetylation inhibitor Psammaplin A (PsA) on the development of bovine aging oocytes in vitro,oocytes were randomly divided into control group,aging group and 50 mmol/L PsA treated aging group (PsA group).Immunofluorescence staining and JC-1 were used to detect the blastocyst rate of bovine oocytes after parthenogenetic activation,the number of cells in blastocysts,apoptosis,reactive oxygen species (ROS),glutathione (GSH) and mitochondrial membrane potential intensity of embryos.The results showed that the blastocyst rate of the aging group was significantly lower than that of PsA and control groups (P<0.05).The blastocyst rate of PsA group was not significantly different from that of control group (P>0.05).The number of cells in the blastocysts of control group and PsA group were significantly higher than that of aging group (P<0.05).The number of cells in the blastocysts of PsA group was not significantly different from that of control group (P>0.05).The apoptosis rate in aging group was significantly higher than that of control and PsA groups (P<0.05),the apoptosis rate of PsA group was significantly higher than that of control group (P<0.05).The GSH level of MⅡ oocytes in aging group was significantly lower than that of control and PsA groups (P<0.05).There was no significant difference in GSH level between control and PsA groups (P>0.05).The ROS level of the embryos in aging group was significantly higher than that of control and PsA groups (P<0.05).The ROS level in PsA group was significantly higher than that of control group (P<0.05).The mitochondrial membrane potential of early embryos of aging group 4-8 cells was significantly lower than that of control and PsA groups (P<0.05).The mitochondrial membrane potential intensity of control group was significantly higher than that of PsA group (P<0.05).In summary,PsA could effectively delay the aging of bovine oocytes and improve the quality of oocytes.

按照国家及甘肃省技术要求、工作流程进行了兰州市国家级公益林区划落界工作,结果为兰州市国家级公益林本期区划落界132611.07 hm^2,剔除了2013年上报中的16104.46 hm^2,在林地类型、区位、权属等方面更符合国家级公益林的条件要求,从地类、生态区位等方面还分析了国家级公益林的面积结构。
